Magnet Assisted Transfection (MATra)

Recent efforts combining nanotechnology and magnetic properties resulted in the development and commercialisation of magnetic nanoparticles that can be used as carriers for nucleic acids for in vitro transfection and for gene therapy approaches including DNA-based vaccination strategies. The efficiency of intracellular delivery is still a limiting factor for basic cell biological research and also for emerging technologies such as temporary gene silencing based on inhibitory RNA/siRNA. Nanotechnology has resulted in a variety of different nanostructures and especially nanoparticles as carriers in a wide range of new drug delivery systems for conventional drugs, recombinant proteins, vaccines and more recently nucleic acids. It is possible to combine superparamagnetic nanoparticles with magnetic forces to increase, direct and optimise intracellular delivery of biomolecules.

Magnet Assisted Transfection (MATra) is an extremely gentle, easy-to-handle, fast and highly efficient technology to transfect cells in culture. Using this new technique, nucleic acids are in a first step associated with specific magnetic nanoparticles (MagTag®). Exploiting magnetic force the full nucleic acid dose is then drawn towards and delivered into the target cells leading to efficient transfection without disturbing the membrane architecture, without causing chromosomal damage or leaving a hole in the cell membrane like other transfection technologiess.

It enables:

  • Transfection of sophisticated and slowly dividing cells such as neuronal cells and hepatocytes.
  • A non-viral transfection method for cells, that are difficult to transfect
  • High transfection efficiencies using DNA, ODN and siRNA

MATra-A Reagent for 200 ug/1000 ug DNA

MATra-A Reagent contains magnetic nanoparticles (MagTag®), which can be loaded with the nucleic acid of interest. Exploiting magnetic force the full nucleic acid dose is then rapidly drawn towards and delivered into the target cells leading to efficient transfection. MATra-A Reagent can be used for adherent cells; for suspension cells MATra-S immobiliser is required in addition. For the transfection procedure a MATra magnet plate is essential.

All MATra reagents with magnetic beads have to be stored distant (> 10 cm) from strong magnetic fields like the Magnet Plates at all times!!! Otherwise agglutination of the beads may lead to complete loss of transfection function.


MATra-S immobiliser up to 7 million / 35 million cells

Suspension cells have to be made adherent first by incubating them with the reagent MATra-S immobiliser which contains magnetic nanoparticles (MagTag®). Then, either Magnet Assisted Transfection with MATra-A Reagent/MATra-si Reagent or Magnet Assisted Lipofection with MA Lipofection Enhancer and IBAfect or any other commercially available lipofection reagent can be performed. For the transfection procedure a MATra magnet plate is essential.

All MATra reagents with magnetic beads have to be stored distant (> 10 cm) from strong magnetic fields like the Magnet Plates at all times!!! Otherwise agglutination of the beads may lead to complete loss of transfection function.


MA Lipofection Enhancer for 200 ug / 1000 ug DNA

Transfection with common lipidic or polycationic reagents can be enhanced by magnetic assistance (“Magnet Assisted Lipofection”). In this case, the nucleic acid has to be combined with MA Lipofection Enhancer in the presence of a common lipid based or polycationic reagent. The formulation of MA Lipofection Enhancer, which contains magnetic nanoparticles (MagTag®), can be used with IBA’s lipofection reagent IBAfect or any other commercially available lipofection reagent.

The MA Lipofection Enhancer is suitable for adherent cells; for suspension cells MATra-S immobiliser is required in addition. Moreover, the MA Lipofection Enhancer can be used for viral transfection. For the transfection procedure a MATra magnet plate is essential.

MATra-si Reagent

MATra-si Reagent is optimised for siRNA applications. It contains magnetic nanoparticles (MagTag®), which efficiently bind nucleic acids, preferentially siRNA. Exploiting magnetic force the full nucleic acid dose is then rapidly drawn towards and delivered into the target cells leading to efficient transfection.

MATra-si Reagent is suitable for adherent cells; for suspension cells MATra-S immobiliser is required in addition. For the transfection procedure a MATra magnet plate is essential.
