Description: Vangl1 (VANGL planar cell polarity protein 1) is an integral membrane protein that belongs to the tetraspanin family having four transmembrane domains. Tetraspanins form large multimolecular complexes on the cell membrane and are involved in cellular signaling, membrane formation, and migration. Vangl1 is expressed at the plasma membrane of different tissues and play a critical role in establishing planar cell polarity (PCP) in epithelial cells and are required for convergent extension (CE) movements during embryogenesis. It is a highly conserved and have evolved from a single ancestral protein Strabismus/Van Gogh found in Drosophila. Mutations in the vangl1 gene cause NTDs (neural tube defects). Ser/Thr phosphorylation of Vangl1 serves as an effector mediating the ITF healing response of the intestinal mucosa. In colon carcinoma cell lines, Vangl1 interacts with KAI1 (CD82) tumor metastasis suppressor. Vangl1 overexpression increases motility and metastasis, whereas its inhibition decreases motility and metastasis of colon cancer confirming migratory phenotype for Vangl1 in colonic epithelial cells.
