Description: H2AK119ub1 designer mononucleosomes (dNucs) consist of 147 base pairs of DNA wrapped around an octamer of core histone proteins (two each of H2A, H2B, H3.1, and H4) to form a nucleosome, the basic repeating unit of chromatin. The dominant form of ubiquitinated histones are monoubiquitinated H2A and H2B. Ubiquitination of H2A is usually correlated with transcriptional repression, while ubiquitination of H2B is more frequently correlated with transcriptional activation. Histone ubiquitination can be induced by DNA damage, therefore playing an important role in DNA damage response. The 147 bp 601 sequence, identified by Lowary and Widom [1], has high affinity for histone octamers and is useful for nucleosome assembly. The DNA in this nucleosome contains a 5' biotin-TEG group.
