Description: CD63 is a multi-pass, lysosomal membrane protein belonging to the tetraspan (or tetraspanin) transmembrane protein family. The structure of CD63 is comprised of 4 domains spanning the membrane; two unequally sized extracellular loops and two, short domains within the cytoplasm, which are thought to participate in signal transduction for some cell types. CD63 also contains a characteristic lysosomal targeting motif, GYWVM, within its C-terminal sequence. Although the exact function of CD63 has yet to be elucidated, it has been implicated as having a role in growth regulation and to be involved in the initial stages of melanoma tumor progression. Expression of CD63 has been observed in dysplastic nevi, hematopoietic cells, radial growth phase primary, and in tissue macrophages. Studies have suggested a role for CD63 in macrophage infection from CCR5-mediated HIV-1. Loss of CD63 expression has been observed within in a patie4nt suffering from a rare, genetic disorder known as Hermansky-Pudlak syndrome (HPS).
