The binding specificity of this antibody for 4-1BB has been confirmed in ELISA analysis using purified 4-1BB-Ig fusion protein, and immunoprecipitation analysis (Shuford et al, 1997).This antibody is able to block 4-1BBL binding to activated DO.11.10 T cells (Shuford et al, 1997). This antibody reacts with activated T cells, and can costimulate T cell proliferation induced by the 145.2C11 hamster anti- mouse CD3 mAb (Shuford et al, 1997). Equally, when administered to mice, this antibody expands innate immune cells, as well as host T cells (Lin et al, 2010). Administration of this antibody alone induced complete tumor regression in E.G7 tumor-bearing mice (Lin et al, 2010). Binding of this antibody to activated T cells in mice receiving heart allografts led to the rapid rejection of the allograft (Shuford et al, 1997).
This chimeric mouse antibody was made using the variable domain sequences of the original Rat IgG2a format, for improved compatibility with existing reagents, assays and techniques.
