This antibody is capable of inhibiting several cell mediated immune functions. It inhibits proliferative responses of T cells stimulated by soluble antigens, mitogens, or allogeneic cells in the initiation of a culture, but not when added after 48 hours. It successfully blocks the lytic activity of cytotoxic T cells and natural killer cells and the migration of neutrophilic granulocytes (PMID: 6644024). This antibody inhibits phagocytosis by reaction with the C3bi receptor, and stimulates phagocytosis by reverse opsonization (PMID: 3880787). This antibody inhibits aggregation of neutrophils in response phorbol myristate acetate, zymosan-activated plasma, and N-formyl-methionylleucylphenylalanine in a dose dependent manner (PMID: 3995185). 60.3 also inhibits polymorphonuclear leukocyte accumulation and plasma leakage in vivo (PMID: 3539230). 60.3 successfully inhibits neutrophil function in vivo and has the potential to be used as an anti-inflammatory agent (PMID: 3693901). This antibody has application in blocking neutrophil mediated organ damage which occurs after myocardial infarct or hemorrhagic shock. Humanized version of this antibody binds CD18 with a 4-fold less affinity than its parent murine version (PMID: 7937713).
This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2a format, for improved compatibility with existing reagents, assays and techniques.
