Immunogen: The original parent antibody SS scFv was generated by intradermally immunizing BALB/c mice with a vector pcD3CAK1-9 which expresses mesothelin under the control of a CMV promoter. Randomized mutations were introduced in the VH and VL CD3 regions to make C10. C10 has S92G-G93S-Y94H mutations in the light chain.

Application Notes

This antibody is recommended to be used for detection of mesothelin in biological samples. This high affinity anti-mesothelin antibody is derived from its parent antibody SS scFv by incorporating S92G-G93S-Y94H mutations in the light chain. The resulting antibody C10 has a 55 times higher affinity towards mesothelin in an ELISA as compared to its parent antibody. The binding characteristics and affinity of the antibody for mesothelin was determined by surface plasmon resonance (BIAcore assay). C10 binds mesothelin with an affinity of Kd = 0.2 nM. Immunoconjugates with C10 as targeting agent and Pseudomonas exotoxin A (PE38) as immunotoxin can be used for identifying samples containing mesothelin through immunohistochemical assays. The cytotoxic activity of the immunotoxin was also checked on antigen positive A1847 cells (PMID: 10385321, 9435250).
