Application Notes

The enzymatic activity of CD73 causes hydrolysis of extracellular nucleotides into membrane permeable nucleosides. This antibody inhibits soluble placental 5'-NT enzymatic activity and recognizes lymphocyte CD73 in indirect immunofluorescence and immunoprecipitation assays. This antibody was also used in the study of cellular distribution of CD73 (PMID: 2137649). It is reported that 1E9 induces vigorous T cell proliferation in the presence of subatomic doses of phorbol myristate and F(ab')2 goat anti-mouse Ig (PMID: 2137649, 2550543). This antibody was also used to study the expression of CD73 in normal mammary gland and breast carcinoma cells using immunohistochemistry (PMID: 1989648). This antibody was also used to detect CD73 in COS cells transfected with CD73 cDNA using flow cytometry (PMID: 7595232). Flow cytometry analysis also showed that anti-CD73 1E9 mAb recognized CD73 on the human breast cancer cell lines LM3 and MDA-MB-231, but failed to detect the murine CD73 variant expressed on isolated mouse mononuclear hematopoietic cells(PMID: 24043904). Bispecific antibody generated using 1E9 and mAb AD2 is reported to inhibit the enzymatic activity of CD73 as well as promotes internalization of CD73.
