Application Notes

This anti-P-selectin antibody was demonstrated to block the P-selectin-mediated adhesion of human platelets (activated by thrombin) to human neutrophils as well as to inhibit the adhesion to human neutrophils of thrombin-activated rat and rabbit platelets (Mulligan et al., 1992; pmid: 1383277). Furthermore, the administration of PB1.3 in lab animals led to the decrease of lung myeloperoxidase levels, reduced accumulation of neutrophils within the pulmonary interstitial capillaries, diminished adherence of neutrophils to endothelial cells, and greatly reduced hemorrhage (Mulligan et al., 1992; pmid: 1383277). Consequently, this antibody can be recommended for the development of the anti-inflamatory treatments. Additionally, PB1.3 was shown to be able to recognize P-selectin in a platelet preparation via Western blot (Mulligan et al., 1992; pmid: 1383277). Further, this clone was also used to identify the expression of P-selectin on thrombin-activated rat platelets by flow cytometry (Mulligan et al., 1992; pmid: 1383277). Further, the application of the antibody PB1.3 in immunohistochemistry in frozen tissue sections demonstrated that the levels of P-selectin in lung vasculature was rising upon administration of the cobra venom factor (CVF) (Mulligan et al., 1992; pmid: 1383277). Another study demonstrated that the administration of the PB1.3 antibody at the time of reperfusion preserved coronary endothelial nitric oxide release and attenuated myocardial cell necrosis (Lefer et al., 1996; pmid: 8769738). Finally, the same clone was also shown to attenuate lipopolysaccharide-induced acute renal failure in rabbits (Hayashi et al., 2001; pmid: 11287780).
