Application Notes
The original human IgG1 version of this antagonist antibody binds notch 1 with an affinity of Kd= 8 nM. This antibody is capable of efficiently blocking the binding of all 4 ligands (Jag1, Jag2, DLL1 and DLL4) to notch 1. This antibody was also capable of specifically inhibiting Notch 1-dependent transcription. In vivo, this antibody was found to show little to no intestinal toxicity in mice. Hu2E6-62 caused decrease in functional angiogenesis by increasing vascular branching and decreasing vascular function. It inhibited angiogenesis induced by the human lung cancer Calu-6 cells. This antibody can be used to inhibit or reduced the proliferation and growth of tumors (WO2012003472).
