This antibody can recognize PD-L2-Fc in an ELISA. Flow cytometric analysis with whole anti-PD-L2 mAb 24F.10C12 and its Fab version revealed approximately the same mean fluorescence intensity upon binding PD-L2 transfected cells. This antibody is capable of blocking the binding of PD-1-Ig to PD-L2 in a dose-dependent manner and with high avidity. This antibody was also used for the immunohistochemical studies of various tissues including thymus, germinal center, placenta, cardiac tissues, and tumors. Blockade of PD-L2 on dendritic cells results in enhanced T cell proliferation and cytokine production, including that of IFN-gamma and IL-10. Both the whole antibody version and Fab version enhanced T cell activation, showing that PD-L2 functions to inhibit T cell activation (PMID: 12538684). This antibody was used for the study of PD-1 pathway on the activation of naive, memory, and recently activated human CD4+ T cells (PMID: 15598424). Antibody blockade of endothelial cell PD-L2 enhanced endothelial cell costimulation of PHA-activated human CD8+ T cells (PMID: 14579280).
