Application Notes

To characterize the mouse version of this antibody a set of binding assays was preformed on cells from NSCLC tissues from biopsies or pleural effusions. First, ELISA was preformed with this antibody. Then and radioimmunoassay and then finally flow cytometry was preformed (Hellstroem et al, 1986; pmid:3731064). Furthermore an immunohistochemistry experiment was preformed to asses the ability of the mouse version of this antibody to bind cancer tissue. The tissue used for the immunohistochemistry was tumor and normal tissue from humans (Hellstroem et al, 1986; pmid:3731064). Finally immunoprecipitation was preformed on cultured NSCLC cells which had been selected for expressing the antigen defined by a given antibody (Hellstroem et al, 1986; pmid:3731064). The ability of the original mouse version of this antibody to supress the was tested on L-2981 tumors grafted unto nude mice. The result showed a significant decrease in tumor growth or even disappearance (Hellstroem et al, 1986; pmid:3462743). The mouse version of this antibody also shows the ability to lyse L6 positive L-2981 and C-3347 cells in presence of human complement (Hellstroem et al, 1986; pmid:3462743). To research the antigen to which the mouse version of this antibody binds immunofluorescence and flow cytometry was preformed on COS cells transfected with cDNA from the the antigen to which this antibody binds (Marken et al, 1992; pmid:1565644). Furthermore it the same COS cells were then also immunoprecipitated (Marken et al, 1992; pmid:1565644).
