Application Notes
The ability of the scFv version of this antibody to bind to PM-1, PBL and granulocytes was assayed by ELISA and flow cytometry. This confirmed that it only bound PM-1 (CA2682059A1). Futhermore the selectivity of the scFv version of this antibody was tested on human tumor tissues including breast, prostate and kidney tumors from 11-14 different donors. breast, prostate and kidney tumors showed the strongest binding. Then to test the specificty of tumors versus normal tissue a tissue microarray with 13 human breast tumor tissues and to a tissue microarray with 4 normal breast tissues using immunohistochemistry staining was preformed with the scFv version of this antibody. 6/13 tumors showed specific staining and none of the normal tissues showed any staining (CA2682059A1). To capture the antigen to which this antibody binds, immunoprecipitation was preformed with the human version of this antibody (CA2682059A1). Futhermore the immuoprecipitated protein was blotted using the human version of this antibody (CA2682059A1). To evaluate the activity of the human version of this antibody as an inductor of apoptosis the effect of the antibody on BT474 was studied. I did indeed induce apoptosis. (CA2682059A1). By detecting the scFv version of this antibody with a secondary antibody it was determined that PM-1 cells showed some internalization of this antibody (CA2682059A1).
