This agonist antibody is derived from the murine parental anti-TRKB antibody 38B8. The binding of this antibody to TRKB blocks binding of BDNF. The original IgG1 version of this antibody has a KD for human TrkB of 2.54 nM, a KD for mouse TrkB of 1.44 nM, and a KD for rat TrkB of 2.29 nM when measured by Biacore at 25° C. The binding affinity of Fab version of this antibody towards human, mouse and rat TRKB was tested using ELISA. This antibody was reported to show greater efficacy in promoting mouse neuron survival than the parental mouse antibody and BDNF. In vivo treatments with this antibody of mice on high fat diet-induced obesity showed changes in body weight. A single intraperitoneal dose of C2 at 2 mg/kg caused a statistically significant body weight reduction compared to vehicle-treated mice as early as day 2 after dosing. The mean maximum weight reduction by C2 antibody was 29.05% by day 14 when the animals were euthanized (US20100196390). This antibody may also find potential application as a therapeutic for Huntington's disease. The original parental mouse was used in one such study as an agonist to promote neuronal cell survival by activating the receptor and down-stream signaling proteins (PMID: 24503862).
This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
