The specificity of the antibody for glycoprotein E was confirmed by ELISA analysis. The antibody detected glycoprotein E by western blot analysis (Niedrig et al., 1994; PMID: 7817895). Neutralization of TBVE by the original format of the antibody and Fab fragment was determined, the EC50 values were respectively 0.24 +/- 0.03 and 35.0 +/- 2.5 µg ml-1. The cryo-EM structure of the Fab fragment in complex with Tick-borne encephalitis virus was determined. A fusion-from-without assay using C6/36 cells was performed to determine if the original format of the antibody and the Fab can prevent membrane fusion in vivo. Results showed that the native TBEV induced cell fusion at low pH, the virus in complex with the original format of the antibody lost this ability, and the virus in complex with the Fab induced cell fusion with lower efficiency than the native virus. The antibody has therapeutic potential (Fuzik et al. 2018; PMID: 29382836).
