This reformatted human antibody was made using the variable domain sequences of the original Human IgG format for improved compatibility with existing reagents assays and techniques.
The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody showed neutralizing activity against TBEV RVPs (IC50 <1 ng/ml) and authentic TBEV Hypr strain (a TBEVWE isolate; IC50=67.2 ng/ml). Immunofluorescence was performed on PS cells using this antibody. Prophylaxis and therapeutic experiments in BALB/c mice infected with TBEV Hypr showed the antibody was efficacious in prevention (even at low doses ~0.005 mg/kg) and early treatment of the infection (up to 3 days post-infection). The crystal structure of the Fab fragment in complex with the EDIII domains from all three TBEV subtypes was determined (Agudelo et al. 2021; PMID: 33831141).
