The in vitro binding characterization of this antibody was done using ELISA and flow cytometry. This antibody can recognize TEM1-expressed on mammalian cells, like human or murine cell lines expressing TEM1 endogenously (HEK-293T, MOV1 and 2H11) or stably transduced with pHRSINTEM1 (hTEM1-MS1, hTEM1-H5V and hTEM1-SKOV3) in a flow cytometric assay. This antibody binds specifically to Endosialin/TEM1-expressing ovarian tumor in vivo, with functional stability over 48 hrs. (PMID: 20837020). The original scFv version of this antibody binds human TEM-1 with a dissociation constant of Kd= 2 nM (PMID: 29313073).The scFv78-saporin immunoconjugate generated using this antibody exerted dose-dependent cytotoxicity with high specificity to TEM1-positive cells in vitro (PMID: 27933426). This antibody was used in the generation of a multivalent Fc-fusion protein called 78Fc, which has desirable characteristics for immuno-imaging applications. The NIR imaging and tomography results suggest that the 78Fc-based NIR tracer performs well in distinguishing mouse- or human-TEM1 expressing tumor grafts from normal organs and control grafts in vivo (PMID: 25051365). This scFv78Fc fusion protein was conjugated to monomethyl auristatin E (MMAE) and saporin to develop potent TEM1-targeting immunotoxins, which showed TEM1-specific toxicity in vitro and in preclinical animal models (Chunsheng et al.; 29313073). The scFv78-Fc was also conjugated with the chelator p-SCN-Bn-CHX-A"-DTPA, followed by labeling with indium-111. [111In]CHX-DTPA-scFv78-Fc binds specifically to endosialin/TEM1 in vitro and in vivo (PMID: 32198704).
This full-length chimeric rabbit antibody was made using the variable domain sequences of the original Mouse scFv format for improved compatibility with existing reagents assays and techniques.
