The specificity of the original format of the antibody was confirmed by ELISA analysis (Lescar et al., 1993; PMID: 7680133). The structure of the Fab fragment in complex with Guinea-Fowl Lisozyme was determined (Lescar et al., 1995; PMID: 7629116). Homogeneous kinetics of Fab and Fv fragments were studied and the KD values were 0.57 +/- 0.05 nM for scFv and 1.6 +/- 0.2 nM for Fab. Since the clone and HyHEL-10 recognized a common epitope, equilibria and kinetics of the two clones were evaluated. Results showed that the clone had greater specificity than HyHEL-10 despite its lower affinity (Pons et al., 2002; PMID: 12237453). The ability of the antibody to modulate in vitro the presentation of hen egg-white lysozyme (HEL) to specific T cells was evaluated, results showed the antibody was able to enhance the HEL presentation to L3E10 T cells (Guermonprez et al., 1999).
