Application Notes

The antibody reacted with purified carcinoembryonic antigen in solid-phase radioimmunoassay and by Western Blot analysis. The antibody was tested by fluorescence-activated cell sorting for cell surface binding to the LS174T WiDr and HT-29 colon carcinoma cell lines. The antibody was tested by FACS for binding to the surface of normal human polymorphonuclear leukocytes where it appeared nonreactive (Muraro et al., 1985; PMID: 2413997). The antibody was shown to bind CEA with a Ka of 1.4 x 10^9 M^-1 (Kuroki et al., 1989; PMID: 2474516). The antibody was used for immunohistochemistry on colorectal, gastric, and lung adenocarcinoma tumor tissues. In addition, pancreatic, breast, gallbladder, bile duct, malignant ovarian teratoma, mucinous endometrial carcinoma, colon cancer, remote colon mucosa and bladder cancer stained positive (Shi et al., 1994; PMID: 7520463 and Robbins et al., 1993; PMID: 8386136). The antibody reacted by immunofluorescence with a mouse colon adenocarcinoma MC-38 expressing high levels of cell-surface CEA cell line, but not with the NCA or BGP transfectants (Robbins et al., 1993; PMID: 8386136). 125I-labeled COL-1 IgG was shown to efficiently and specifically target the LS-174T human colon carcinoma xenograft in athymic mice. 131I-labeled COL-1 demonstrated a reduction of tumor growth rate in the same model (Siler et al., PMID: 8292968). I131-labeled COL-1 was tested in phase I trial in patients with gastrointestinal malignancies (Yu et al. 1996). The humanized version of the antibody was constructed and used in the detection and treatment of a CEA-expressing tumor (US8828717B2).
