Application Notes
The antibody is a humanized version of mab23. The antibody had an activity EC50 of 0.34 µg/ml for activating human T cells to release IL-2. Further, the antibody activates human DC cell to stimulate human T cell activity (EC50 of 0.21nM), as shown by MLR assay. The humanized antibody efficacy in vivo was evaluated of the present invention. Tumors treated in mice showed an inhibition in tumor growth when treated with a combination of the antibody and PD-1 antibody. The binding affinity of the antibody to hLAG-3 was measured by surface plasmon resonance (Kd= 0.2 nM). The antibody showed a good blocking activity. The antibody was used in combination with an anti PD-1 antibody (Nivo or Pem) into a bispecific antibody (CN112079925).
