The binding affinity of the antibody to PD-L1 was measured by surface plasmon resonance. The binding affinity of the antibody was measured on the cell lines with high expression of PD-L1 by FACS. The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody had good activity in blocking the binding of PD-L1 to PD-1 and PD-L1 to B7-1 is shown by ELISA analysis. The antibody could block the binding of PD-1 and PD-L1 at the cellular level in in vitro experiments. The antibody was used for in vivo experiments on colon cancer tumor model. B-hPD-1 mice with human PD-1 and MC38-hPD-L1 colon cancer cells with human PD-L1 were treated with the antibody. Results showed inhibition of tumor volume growth in mice treated with the antibody.
This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
