The original version of this antibody (human IgG1) was characterized using ELISA, SPR, and FACS assays to isolate it and assess its binding specificity and affinity. It bound its target with a Kd of 120 nM. In a FACS-based in vitro neutralization assay, this antibody exhibited a relatively low neutralization potency compared to its Gn-specific antibody counterparts with a median inhibitory concentration (IC50) of 1,370+/-530 ng/ml. However, it can still prevent RVFV infection at high concentrations. In an in vivo mouse model, mice treated with this antibody showed lower survival rates compared to those treated with Gn-specific antibodies. However, this antibody still contributed to clearing the virus, as evidenced by the survival of one mouse in the respective group, indicating that this antibody can neutralize RVFV infection at high doses also in vivo (Wang et al., 2019; PMID: 30936489).
This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
