The antibody showed neutralizing activity against SFTSV infection in Vero cells (Guo et al. 2013; PMID: 23863504). To verify neutralizing activity, full-length human IgG1 antibody was constructed. The recombinant antibodies were expressed by 293T mammalian cells and the concentration required to obtain 50% neutralization of 100 TCID50 SFTSV in vitro was 44.2 µg/mL. The interaction between Gn and the antibody was further demonstrated by surface plasmon resonance (SPR) assays with a dissociation constant (Kd) of 25.9 nM. The crystal structure of SFTSV Gn head domain in complex with the Fab fragment of the antibody was determined. The antibody could recognize the denatured SFTSV Gn head by WB (Wu et al., 2017; PMID: 28827346).
This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
