Application Notes
The antibody was shown to completely block β-catenin signaling, demonstrating it is a specific inhibitor of RSPO2. The antibody blocked soluble RSPO2 binding to LGR5. The antibody had a binding affinity for human RSPO2 of 0.14 nM and mouse RSPO2 of 0.35 nM. The antibody demonstrated strong anti-tumor growth activity in combination with gemcitabine in a pancreatic xenograft model and inhibition of pancreatic tumor growth in vivo as a single agent and in combination with additional therapeutic agents. The humanized version of the antibody was produced. The humanized version had a binding affinity for human RSPO2 of 0.13 nM (US8802097B2).
