This nanobody was shown to detect primary tumors and metastatic sites in vivo by specifically binding to the ECM with excellent specificity in multiple models of breast cancer, including human and mouse triple-negative breast cancer, and in melanoma, as measured by PET/CT imaging with 64Cu-NJB2. This nanobody recognized EIIIB-His by ELISA and in FN-containing EIIIB immunohistochemistry (IHC). Furthermore, its binding affinity to immobilized single EIIIB domain and a large recombinant rat FN fragment comprising FNIII7-15, including EIIIB (FN 7-15 EIIIB), was 1.9 x 10-9 M and 2.25 x 10-9 M, respectively, as determined by biolayer interferometry (BLI) (Jailkhani et al., 2019; PMID: 31068469). This nanobody was used for IHC of B16F10 and MC38 tumors to detect EIIIB expression (Xie et al., 2019; PMID: 30936321).
