The reactivity of the original antibody (alpaca VHH) against norovirus VLPs and P domains was determined using ELISA. It was shown to be reactive with antigenically distinct norovirus VLPs, including GII.4, GII.10, and GII.12. Using isothermal titration calorimetry, a Kd of 7 nM was measured against GII.10 P domain and a Kd of 3.5 nM against GII.4 P domain. The crystal structure of this antibody in complex with norovirus GII.10 P domain, Saga-2006 GII.4 P domain, and 2012 NSW GII.4 P domain were determined. This antibody can bind intact norovirus VLPs and virions and promotes their disassembly (Koromyslova & Hansman, 2015; PMID: 25520510). The epitope of this antibody was further characterized in X-ray crystallography and cryo-EM. This antibody's binding mechanism involves impairing capsid motions and integrity, which inhibits multiple stages of the Norovirus life cycle (Koromyslova & Hansman, 2017; PMID: 29095961).
