The binding specificity of the original camelid (alpaca) single-domain antibody (sdAb; VHH) was tested in an ELISA. This antibody was used to generate an anti-avian influenza nucleoprotein and anti-human IgA bi-specific construct together with Ab04649 [A8]. The bi-specific construct detected a range of influenza virus strains in ELISA, including low-virulence and vaccine strains. Namely, H1N1, H3N2, H5N3, H5N1 (LP), H7N3(LP), H8N4, H10N8, H11N6, H12N5, H2N2, H4N6, H5N9, H6N2, H7N7, H9N2, H10N7, H12N6, and H13N6. The bi-specific construct also exhibited a significantly prolonged stability in vivo in Balb/c mice compared to the sdAb version of the antibody (CN111116752B).
