Application Notes
The binding affinity of the original format of the antibody to CD8 was measured by surface plasmon resonance (Kd= 0.9 nM). FACS analysis of splenocytes and lymph nodes gated on CD45+CD19-CD3+ cells, confirming that the original format of the antibody stained CD8+ cells. The antibody competed with a commercial anti-CD8α antibody, showing it bound to the CD8α chain. 89Zr-PEGylated antibody was used for PET imaging on mice, to track the presence of intratumoral CD8+ T cells (Rashidian et al., 2017; PMID: 28666979).
