The original human IgM antibody's reactivity was confirmed through various immunoassays, including immune adherence (IA) and immune adherence absorption (IAA) assays. It exhibited strong binding to gangliosides and effectively detected a range of human tumor tissues, such as melanoma, colon adenocarcinomas, and lung adenocarcinomas. The antibody demonstrated significant cytotoxic activity against antigen-positive tumor cells in the presence of complement. It showed notable reactivity with melanoma (62% positive) and lung cancer (100% positive) while showing less reactivity with other cancers. Neuraminidase treatment effectively inhibited its binding, affirming its specificity for gangliosides. The antibody has been used both therapeutically and diagnostically, showing efficacy in treating cutaneous malignant melanoma through intralesional injection, resulting in tumor necrosis and regression. It has been used to produce hybridomas that generate anti-idiotypic antibodies (anti-ids); beta-type anti-ids inhibit the antibody's binding and are useful for treatment and immunomodulation, while alpha-type anti-ids are used in diagnostic assays. Diagnostic applications include a three-step cell-ELISA and immunoperoxidase staining, both of which effectively detect GM3 antigens on tumor cells and tissue sections (US5419904A).
