Application Notes

This antibody was used for the development of a H3K27me3-specific intracellular antibody or H3K27me3-mintbody, which was a fusion protein comprising variable domain regions of 2E12LI antibody fused to a fluorescent protein and expressed in mouse teratocarcinoma cell line MC12. This mintbody was capable of tracking H3K27me3 in living cells. Mouse MC12 cells, which stably express H3K27me3-mintbody (sfGFP) was used to localize H3K27me3-enriched chromatin in living cells. The binding specificity of bacterially expressed and purified 2E12LI-sfGFP in vitro was evaluated using a modified histone peptide array. (PMID: 33605056).
