This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
The original version of this antibody was used in a radioimmunoassay (RIA), a mixed Hemadsorption Assay (MHA), and an immunofluorescence (IF) assays to validate its specificity for human colorectal carcinoma cells. It was also tested on fresh tumor tissue from colorectal carcinoma patients, where it was found to bind selectively to tumor cells, but not normal intestinal tissue, further confirming its specificity for colorectal carcinoma (Herlyn et al., 1979; PMID: 286328). This antibody was crystallized with and without its target antigen. Furthermore, in a surface plasmon resonance (SPR) assay a KD of 14.7 µM was measured, and the antigen recognition of an scFv version of this antibody was measured via flow cytometry (FC) (Borenstein-Katz et al., 2021; PMID: 34119488). The use of this antibody in IHC was successfully demonstrated in a doctoral dissertation (Khilji, 2023).
