This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2b format, for improved compatibility with existing reagents, assays and techniques.
The original format of the antibody bound to cell surface CD20, such as human blood lymphocytes and the Raji B lymphoblastoid cell line, by immunofluorescence analysis. The original format of the antibody bound to primary B cells and B lymphoma cell lines expressing CD20 in indirect immunofluorescence staining assays (US9416187B2). The IgG1 and IgG1 YTE formats of the antibody were able to efficiently elicit ADCC with an IC50 of 1.3 +/- 0.5 and 3.7 +/- 1.4 ng/mL, respectively with Daudi as target cells and 4.6 +/- 1.6 and 5.2 +/- 1.7 ng/mL with Raji cells. Further, the IgG1 and IgG1 YTE formats of the antibody were able to potently elicit CDC with IC50 values of 34 and 160 ng/mL, respectively (Borrok et al., 2017; PMID: 28057542).
