The specificity of the original format of the antibody was confirmed by ELISA analysis. The binding affinity of the original format of the antibody to HER2 was measured by surface plasmon resonance (Kd= 8.5 nM). The original format of the antibody was well tolerated in in vivo experiments with cynomolgus monkeys (Adams et al., 2005; PMID: 16151804). The original format of the antibody inhibited SK-BR-3 relative cell proliferation by about 20%. The original format of the antibody inhibited the generation of a HRG-induced tyrosine phosphorylation signal at Mr 180,000 (IC50= 2,8 nM). The antibody inhibited HRG binding to MCF7. The antibody was able to inhibit cell proliferation of MDA-MB-175 cells. The antibody blocked heregulin dependent association of ErbB3 with ErbB2 in both MCF7 and SK-BR-3 cells. The original format of the antibody was used for the treatment of the Calu-3 tumor-bearing mice, and it significantly inhibited the growth of the tumors. Furthermore, the combination of HERCEPTIN® and the original format of the antibody was superior to either monoclonal antibody administered alone. The original format of the antibody suppressed growth of colorectal xenografts and inhibited breast cancer tumor growth in vivo (US7862817B2).
