Short Description

This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2b format, for improved compatibility with existing reagents, assays and techniques.

Application Notes

This antibody was used for an ELISA on human C3d. This antibody was used for a western blot on human C3d, it showed strong binding. The affinity of this antibody to C3d was determined using surface plasmon resonance, it showed strong binding with a Kds of 0.47 nM. This antibody also showed it could inhibit the CR2-C3d interaction in a competition ELISA. This antibody was used in vivo on mice, it was shown that this antibody could detect tissue- bound C3 fragments, it readily detected C3 along the glomerular basement membrane (Thurman et al., 2013; PMID:23619360). The structure of this antibody was determined using x-ray crystallography. A fusion protein of this antibody with the first 10 consensus repeats of the human CR1 protein was used in vitro for a Wieslab assay, a hemolysis assay and a HMEC assay. The pharmacokinetics (PK) in mice were determined for various fusion proteins containing this antibody (Fahnoe et al., 2022; PMID:35784298). A fusion protein of this antibody with factor H was used for a complement inhibition assay it showed an IC50 of 73 nM. This antibody was used for immunofluorescence on human skin biopsies and kidney tissues (Liu et al., 2024; PMID:38382529).
