Short Description

This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG4 format, for improved compatibility with existing reagents, assays and techniques.

Application Notes

The original antibody was used to block PtdSer binding to TIM3. Jurkat E6-1 cells were treated with paclitaxel to induce apoptosis and promote the accumulation of PtdSer on the cell surface. Then, it was mixed with mFc-labelled TIM3, this antibody, and control antibody. The TIM3 protein bound to PtdSer on the surface of apoptotic cells was detected with PE-labelled anti-mouse Fc antibody using flow cytometry. It showed that this antibody could inhibit binding of TIM3 to PtdSer in a dose-dependent manner.The affinity of this antibody to TIM3 was determined using surface plasmon resonance, it showed a affinity of 3.64*10^-5 Kd . The functional activity of this antibody was determined by measuring IFN-γ secretion by DC-T cell co-culture assay, it showed that it could increase secretion of T-cell IFN-y (CN112062850B).
