This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.
This antibody was reported to mediate AQP4-directed antibody-dependent cellular cytotoxicity and complement-mediated lysis in vitro. The binding characterization of this antibody to AQP4 expressed on Human Fetal Astrocyte (HFA) was done by quantitative flow cytometry (PMID: 19938104). Binding of rAb-58 to the M1 and M23 AQP4 isoforms was measured by the fluorescence ratio imaging method (PMID: 21454592). CHO cells expressing M1- or M23-AQP4 when labeled with rAb-58 conjugated to the red fluorophore Cy3 (rAb-58-Cy3), showed rapid internalization of M23-AQP4 than M1-AQP4 when expressed separately in transfected cells. When both isoforms were co-expressed in OAPs in transfected cells, the antibody causes simultaneous cointernalization of M1- and M23-AQP4 isoforms (PMID: 22987455).
