Short Description

This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgM format, for improved compatibility with existing reagents, assays and techniques.

Application Notes

The original antibody format (Mouse IgM) was generated and evaluated. Its specificity was determined by indirect whole-cell and purified polysaccharide ELISA. It exhibited strong binding to serogroup B capsular polysaccharide (OD₄₀₅ ~ 0.85 on strain 99M) with only background-level reactivity to serogroup C and the other 12 serogroups, demonstrating exquisite type B specificity (Berry et al., 2005; PMID: 15589322). This antibody was successfully used to investigate capsule expression in Neisseria meningitidis serogroup B. It was used in a whole-cell ELISA to quantify total capsular polysaccharide production, in flow cytometry to detect surface and intracellular capsular polysaccharide, and in immunogold electron microscopy to visualize capsular polysaccharide localization (Tzeng et al., 2005; PMID: 15731047). This antibody underwent genetic analysis to reveal its unmutated V genes, structural modeling to identify a shallow combining site groove, and used as a control for autoreactivity to confirm strong PSA binding, contrasting with non-autoreactive anti-N-Pr MBPS mAbs (Moe et al., 2006; PMID: 16140379).
