Short Description

This is a reformatted human IgG1 Fc Silent Fc Silent™ antibody, based on the original human IgG1 format, created for improved compatibility with existing reagents, assays and techniques.

Application Notes

The binding reactivity of this antibody to various sdAbs was determined using ELISA. The binding of this antibody to the VHH did not disrupt VHH/antigen interaction, and so it is appropriate for secondary labeling to assess cellular or tissue reactivity of VHH molecules. The binding ability of this antibody to detect surface expressed sdAb-based CAR constructs was also tested. This antibody could detect Jurkat T cells stably expressing sdCD22 CAR constructs. It was reported that a 1 :1 mix of antibodies 2A3 and 3H12 provides the best overall breadth of binding to diverse VHH proteins. This antibody was also used for the labelling and staining of frozen human tissues using immunohistochemistry. The tissues were first stained with CD22 specific nanobody and the anti-VHH antibody 2A3 was used a secondary antibody (WO2024164074). This antibody potently blocked VHH-CAR-T activation and cytolytic killing of target cells. When immobilized, anti-VHH antibodies induced strong activation and expansion of VHH CAR-T cells (McComb et al. 2025).
