TM4 cells(black) were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with TNF Antibody (bs-2941R) at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green) and isotype control (orange).

Lane 1: Rat Cerebrum lysates probed with TNF Receptor I Polyclonal Antibody, Unconjugated (bs-2941R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37°C.

THP-1 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1%PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. L1 mouse intestine lysates probed with Anti TNFR1/TNF Receptor I Polyclonal Antibody, Unconjugated (bs-2941R) at 1:200 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000 for 90 min at 37˚C. Predicted band 40kD/26kD. Observed band size:40kD/26kD.

Description
Description: Receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Contributes to the induction of non-cytocidal TNF effects including anti-viral state and activation of the acid sphingomyelinase.
