Description: Macrophages function in phagocytosis of foreign organisms and materials as well as resistance to tumors1. Additionally, they play roles in T lymphocyte immune responsiveness, inflammatory responses, and regulation of hematopoiesis. Lysosomes are the membrane-bound vacuoles that digest foreign materials and perform specialized autolytic functions2. Lysosomes also play a role in antigen presentation, cholesterol metabolism, and hydrolysis of exogenous materials3 . CD107b (LAMP-2, Mac-3) is a member of the lysosome-associated membrane protein (LAMP) family and is an important component of the lysosomal membrane involved in autophagy3,4 . CD107b is a highly glycosylated transmembrane protein, with glycosylation being necessary for its stability5,6. Mice deficient in CD107b have a severe phenotype, with 50% dying aged 20-40 days6. In humans, mutations in CD107b lead to Danon disease. The immunogen used to generate M3/84 was produced by first purifying surface glycoproteins from macrophage membranes of peritoneal exudate cells of C57BL/6 mice5. Next, previously recognized antigens were removed by immunoadsorbent columns to obtain a glycoprotein- enriched fraction. Then, (Lewis x BN) F1 rats were injected and splenocytes fused to mouse myeloma NS1 for hybridoma production.
