Description: Folates represent an important role in protein and nucleic acid biosynthesis and are recognized in particular for their role in spinal canal and brain development during early pregnancy. Recently, Yamaguchi et al, 2007 have shown that Folate receptor 4 (FR4), a subtype of the receptor for the vitamin folic acid is expressed at high levels in both natural and TGF-b-induced Treg cells. A majority of the CD4+ Tregs express FR4 along with CD25 and FoxP3. In addition, there are functional similarities in regulating immune response by FoxP3 and FR4 expressing CD4+ Tregs which express CD25 as well. CD4+CD25+FR4hi T cell population may be used as a functional immuno-suppressor Treg population which should express FoxP3. FR4 may be considered as a cell surface marker for Tregs. The antibodies produced by clones 11E196 and 11E197 recognize FR4 in Tregs. FR4 is also known as folate receptor delta and folate-binding protein 3. The FR4 antibodies in combination with CD4 and CD25 antibodies may be used for isolation (both FACS and magnetic beads sorting) and staining of Tregs using simple surface staining while keeping the Tregs viable for subsequent down stream applications. FR4 is known to be expressed exclusively in lymphoid tissues.
