Description: Murine (m) 1B4 was humanized (h) by the CDR-grafting method to create an anti-CD18 antibody with similar avidity, specificity, and efficacy as the murine antibody. In rhesus monkeys, h1B4 is less immunogenic than m1B4 and elicits only anti-idiotypic antibodies. Additionally, h1B4 and m1B4 exhibit equivalent staining specificity and intensity in rabbit bone marrow leukocytes. For humanization, human VH and VL FR were chosen based on high identity with their corresponding m1B4 FR3. Once grafted with the m1B4 CDR, the human VH and VL FR were expressed as fusion proteins with the IgG4 and Cκ human constant regions, respectively. The h1B4 monoclonal antibody has been used in multiple studies of adhesion in different model systems and to measure the expression of CD18 in B-ALL cell lines.
