Suitable for use in Western Blot, Immunoprecipitation. Western Blot Analysis: 0.5-2.0 ug/ml detects p70 S6 kinase in RIPA lysates from HEK293, PC-12, and A431 cells. HEK293 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-p70 S6 kinase (2 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 4 ug immunoprecipitates p70 S6 kinase from 500 ug of HEK293 RIPA lysate. Immunoprecipitation Kinase Assay: 5 ug immunoprecipitates active p70 S6 kinase from IGF-1 stimulated (15nM) human HEK293 cell lysate containing 1 uM microcystin. The assay was performed using a S6 Kinase Assay Kit, which uses specific substrate peptide as a kinase substrate and contains a separate inhibitor cocktail which blocks the activity of other serine/threonine kinases such as protein kinase A, protein kinase C, and calmodulin dependent kinases.
