Application Notes

Starting trial dose for making mice thrombopenic: We recommend diluting antisera 1:10 in saline and filtering through a 0.2um filter. Give 0.5ml of this diluted, filtered antisera i.p. per 10g of mouse. This must be repeated every 2-3 days to keep cell counts low.This polyclonal hyperimmune antisera can kill injected mice when used in very high doses. The recommended doses given here are for beginning trials to be used on 1 to 3 animals. They are to help the researcher achieve the correct dose for a particular experiment. After the researcher believes he/she has the correct dose, the researcher should plan the experiments using the same lot of antisera. If this is not done prior to the study, the animals can be easily killed by overdosing with this hyperimmune sera. The antibodies can clump circulating cells which can lodge in the lung and kill the animal.
