Application Notes

Human monocyte derived Dendritic cells were prepared in 8 day culture. Five x 105 cells per tube were washed and incubated 45 minutes on ice with 80 ul of antihuman CD273/Biotin at concentration of 10 ug/ml. They were then washed twice and incubated with Streptavidin/R-PE, after which they were washed twice, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.39 log10 fluorescent units when compared to isotype control Mouse IgG2a/Biotin. Binding was blocked when reagent was pre incubated with a 10-fold excess of recombinant CD273-muIg. The applications listed have been tested for the unconjugated form of this product. Other forms have not been tested.
