Suitable for use in ELISA, Western Blot, Neutralization. Neutralization of Feline CCL5 Bioactivity: The exact concentration of antibody required to neutralize rfeRANTES activity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when that cytokine is present at a concentration just high enough to elicit a maximum response. The ND50 for this lot of anti-feline RANTES antibody was determined to be approximately 0.5-2.5 ug/ml in the presence of 80 ng/mL of rfeRANTES, using transfected BaF/3 cells in a chemotaxis assay. The specific conditions are described in the figure legends. Western Blot: 0.1-0.2 ug/ml with the appropriate secondary reagents to detect feline RANTES. The detection limit for rfeRANTES is approximately 1 ng/lane under non-reducing and reducing conditions. Direct ELISA: 0.5-1.0 ug/ml with the appropriate secondary reagents to detect feline RANTES. The detection limit for rfeRANTES is approximately 0.5 ng/well. In this format, this antibody shows less than 5% cross-reactivity with rcrRANTES, rhRANTES, and rmRANTES.
