Application Notes
Immunoblotting, Immunohistochemistry: Immunofluorescence, Immunoprecipitation, Test at 1-10ug/ml in all applications. Sandwich ELISA as capture or HRPconjugated detection antibody. These are recommended concentrations; enduser should determine optimal concentrations for their applications. 96-well ELISA plates are coated with capture MAb at 2.5-5ug/well and incubated at 4oC overnight. The next day 300ul of blocking buffer is added, and plates are again incubated at 4 oC overnight. The next day plates are washed in PBS and serial dilution of Aβ samples are added; plates are incubated overnight at 4oC. The next day plates are washed in PBS, and HRP-conjugated detection Aβ MAb is added; plates are incubated for 4hrs at room temperature. Plates are washed in PBS-Tween and developed with TMB substrate for 5 mins. Antibody has only been tested in IHC and IF with formalin-fixed, paraffin-embedded samples.
