Description: The Cas9/guide RNA (Cas9/gRNA) system is commonly used for genome editing. The gRNA of the system recognizes its target sequence in the genome, and the Cas9 nuclease acts as a pair of scissors to cleave the double strands of DNA. The cap0 structure is added to the Cas9 mRNA by co-transcriptional incorporation of Anti-Reverse Cap Analog (ARCA). The capping of ARCA ensures high translation efficacy, while the addition of 5-moUTP (5-Methoxy-UTP) reduces the host cell immune response and increases mRNA stability.
Cas9 mRNA with Cap 0 structure, modified by 5-moUTP, providing higher transcription efficiency and suppressing RNA-mediated innate immune activation.
