Test Procedures

1. Sample collection and preparation.

2. Keep the kit and sample to room temperature prior to testing.

3. Add sample to the sample well using a micropipette.

4. Add buffer to the sample well.

5. Wait for the colored line(s) to appear and read the result at 10 minutes .

Interpretations of Results

IgM Positive: indicated of primary COVID 19 infection.

IgG and IgM Positive: indicated of secondary COVID 19 infection.

IgG Positive: indicated of past COVID 19 infection.

Negative: indicated of no previous infection.

Invalid: indicating that the experiment is failed.

Proteinase K
Proteinase K is a kind of high activity proteinase of subtilisin, which is used to degrade proteins in biological samples. It can be used to digest various proteins, and it can be used in a variety of molecular biology, cell biology and other related experiments, such as genomic DNA extraction, enzyme digestion and removal, cell permeability and so on.
Proteinase K is mainly used for the production of nucleic acid extraction kit, especially for nucleic acid extraction from samples of nucleic acid diagnosis and detection in vitro diagnosis (IVD) industry.

Shortage of supply

Activity result
Elabscience® standard production process requires that the activity ratio ≥30 U/mg. In fact, the activity ratio exceeded the expected production standard, content the application requirements of customers.
Unit definition: One unit is defined as the amount of enzyme that catalyzes the formation of 1 μmol of tyrosine per minute at pH 7.5 at 37°C.

Calibration curve
According to the absorbance A275 for the vertical axis, the different concentrations of tyrosine as the horizontal axis, the standard curve was drawn.

Detection of DNase and RNase residues
Use DNase and RNase detection kit to detect the DNase and RNase residues with different Lots of Proteinase K. The results showed that there were no DNase nor RNase in Proteinase K.

The study shows that the transmission capacity of the disease increases by about 50%, but there is no evidence to prove the increase of pathogenicity. E484k is found in RBD region, which is considered to be the main reason for the decrease of neutralization activity of neutralization antibody.

Also known as CAL.20C, researchers at cedars Sinai Medical Center first observed it in California in July 2020. Recently, it has appeared in other countries, such as Australia. It was found in the UK first, and the research shows that its communication power has increased by more than 30%. Davies team modeled the data from three regions of the UK and estimated that the transmissibility of the mutant was 56% higher than that of the previous mutants. Spike Protein

The spike protein is a large type I transmembrane protein containing two subunits, S1 and S2. S1 mainly contains a receptor binding domain (RBD), which is responsible for recognizing the cell surface receptor, and S2 contains basic elements needed for the membrane fusion.

Nucleocapsid Protein (N protein)

The coronavirus N protein is abundantly produced within infected cells. N protein has multiple functions, including binding to viral RNA to form the ribonucleocapsid and has also been proposed to have roles in virus replication, transcription and translation.In host cells, N proteins have been shown to cause deregulation of the cell-cycle, inhibit the production of interferon, up-regulate the production of COX2, up-regulate the activity of AP1, and induce apoptosis in serum deprived cells-of all which may have possible pathological consequences.

Membrane and Unstructural Proteins

Membrane protein and Envelope protein are the key to the efficient assembly of coronavirus.
3C-like proteinase of SARS-CoV-2 virus is a key protein in RNA replication.
Papain-like protease (NSP3) plays an important role in the precursor cleavage of various viral proteins to form a series of functional proteins needed in viral replication.
3C-like proteinase and Papain-like protease are important targets for the development of antiviral drugs, which can inhibit virus replication and reduce related mortality.

ACE2 Protein

ACE2 (Angiotensin Converting Enzyme 2) is the receptor of SARS-CoV-2 infected cells, and the infection process depends on the serine protease TMPRSS2. Blocking the interaction between S protein and ACE2 is an effective way to treat coronavirus infection.
